چکیده
Abstract While some individuals infected by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) present mild-to-severe disease, many SARS-CoV-2-infected are asymptomatic. We sought to identify the distinction of immune response between asymptomatic and moderate patients. performed single-cell transcriptome T-cell/B-cell receptor (TCR/BCR) sequencing in 37 longitudinal collected peripheral blood mononuclear cell samples from asymptomatic, moderate, patients with healthy controls. Asymptomatic displayed increased CD56 bri CD16 ? natural killer (NK) cells upregulation interferon-gamma effector CD4 + CD8 T NK cells. They showed more robust TCR clonal expansion, especially cells, but lack strong BCR expansion compared Moreover, have lower interferon-stimulated genes (ISGs) expression general large interpatient variability, whereas various magnitude temporal dynamics ISGs across multiple populations than a patient disease. Our data provide evidence different signatures SARS-CoV-2 infections.

Single cell transcriptional analysis reveals novel innate immune cell types
Single-cell analysis has the potential to provide us with a host of new knowledge about biological systems, but it comes with the challenge of correctly interpreting the biological information. While emerging techniques have made it possible to measure inter-cellular variability at the transcriptome level, no consensus yet exists on the most appropriate method of data analysis of such single ce...
متن کاملTranscriptomic profiling of microglia reveals signatures of cell activation and immune response, during experimental cerebral malaria
Cerebral malaria is a pathology involving inflammation in the brain. There are many immune cell types activated during this process, but there is little information on the response of microglia, in this severe complication. We examined microglia by genome wide transcriptomic analysis in a model of experimental cerebral malaria (ECM), in which C57BL/6 mice are infected with Plasmodium berghei AN...
متن کاملCD8 single-cell gene coexpression reveals three different effector types present at distinct phases of the immune response
To study in vivo CD8 T cell differentiation, we quantified the coexpression of multiple genes in single cells throughout immune responses. After in vitro activation, CD8 T cells rapidly express effector molecules and cease their expression when the antigen is removed. Gene behavior after in vivo activation, in contrast, was quite heterogeneous. Different mRNAs were induced at very different tim...

Description of the technology Understanding a patients’ immune status not only from immune cell phenotyping, but also through analysis of functional signaling capacity, enables the generation of a more comprehensive understanding of the complex mechanisms responsible for immunological tolerance in cancer, and generates data that is complementary to other non-functional phenotypic data sets such...
متن کاملImmune profiling of Alzheimer patients.
Alzheimer's disease (AD) is characterized by extracellular senile plaques in the brain, containing amyloid-β peptide (Aβ). We identify immunological differences between AD patients and age-matched controls greater than those related to age itself. The biggest differences were in the CD4+ rather than the CD8+ T cell compartment resulting in lower proportions of naïve cells, more late-differentia...
